Abstract

Cellulases are enzymes naturally produced by many organisms and used for cellulose hydrolysis. They are well-known for their widely employed industrial application. The recent study was aimed at isolating and screening cellulase producing bacteria from soil samples. Soil sediments were collected from Lake Chitu and transported into Applied Microbiology laboratory.Astu Cellulase producing bacteria were isolated and screened from soil sediments. Various biochemical tests, Protein profile analysis, and molecular characterization were conducted for the identification of these cellulase producing bacteria. Data analyses were conducted using Micro Soft excel, and Origin 2019b software. The sequence analyses were conducted using MEGA 11 version. In this study, a total of 10 cellulase producing bacteria isolates out of 45 isolates were obtained from soil sediments of Lake Chitu. From these isolates, better cellulase producers were identified as Bacillus cereus CPBILCS-9, Bacillus licheniformis CPBILCS-14, Proteus vulgaris CPBILCS-4, and Pseudomonas sp. CPBILCS-13 using matrix assistance lesser desorption time of flight mass spectrometer (MALDI TOF MS). One of the identified cellulase producing bacterial isolates was distinguished as Bacillus tequilensis CPBILCS-15 using 16Sr RNA gene sequence. In this study, it was also noticed that carbon source like glucose, lactose, and sucrose were employed for the growth of Bacillus tequilensis CPBILCS-15 at suitable temperature (37oC) and pH7.5. In conclusion, potential cellulose producing bacterial isolates can be screened from their natural ecosystems and employed for the production of cellulase which used for various industrial applications.
